Abstract
The NISTCHO cell system (RM 8675) provides a standardized Chinese hamster ovary (CHO) cell culture platform for studies of recombinant monoclonal antibody production and bioprocess characterization. Reproducible recovery, expansion, cultivation, and monitoring of NISTCHO cells are important for generating materials with controlled and documented process histories. This Special Publication describes an integrated upstream workflow beginning with recovery of cryopreserved NISTCHO cells and continuing through automated cell concentration and viability measurement, preparation of chemically defined feed media, seed-train expansion, and fed-batch cultivation in a single-use stirred-tank bioreactor. The procedures define operating conditions for temperature, agitation, dissolved oxygen, pH, feeding, glucose supplementation, antifoam addition, sampling, harvest, and initial clarification. A representative 14-day process profile is included to illustrate typical NISTCHO culture behavior. The procedures are intended to facilitate reproducible implementation of the NISTCHO upstream process and generation of cNISTmAb-containing harvest material for downstream processing and analytical studies.
